Internal Control DNA in Polymerase Chain Reaction for Mycobacterium tuberculosis

Abstract

|
In spite of an exquisite sensitivity in detection M. tuberculosis in clinical specimens. PCR suffers from false negative results if the specimens contained DNA polymerase inhibitors of if the test failed due to the technical mistake. For this
reason it
is essential to include the internal control in each PCR tube to rule out this problem.
We selected easily obtainable and well known lambda DNA sequence as an internal control. IS6110 or IS1081 was amplified together with the lambda DNA to detect M. tuberculosis in clinical specimens. It was found that the 100fg of lambda DNA did
not
affect amplification of M. tuberculosis IS6110 DNA, while it significantly decreased amplification of IS1081. As a result the lambda DNA was found an useful internal control DNA when targeted IS6110 for amplification.
